Overview

GRB2-associated and regulator of MAPK protein 1 (GAREM1) is an adaptor protein that functions downstream of the epidermal growth factor receptor (EGFR), regulating activation of the MAPK/ERK signaling pathway[2][5]. It is ubiquitously expressed and plays a role in intracellular signal transduction, contributing to cell proliferation and potentially other cellular processes[1][2][5]. The gene encodes multiple isoforms, and its paralog GAREM2 is brain-specific and involved in neuronal differentiation, while GAREM1 may also have unique roles through nuclear translocation mediated by a specific nuclear localization sequence[1]. No therapeutic drugs, clinical mechanisms of action, or recognized biomarker roles for GAREM1 were reported in the current sources. However, disease association with spondylocarpotarsal synostosis syndrome has been reported, and its involvement in cancer is possible due to its influence on mitogenic pathways[2].
