Overview

GRIP1-associated protein 1 (GRIPAP1, commonly called GRASP-1) is a neuronal guanine nucleotide exchange factor (RasGEF) and prominent scaffold protein involved in the trafficking, recycling, and synaptic localization of AMPA-type glutamate receptors through direct interaction with GRIP1. It regulates the recycling of AMPA receptors to dendritic membranes, essential for maintaining dendritic spine morphology and synaptic plasticity processes such as long-term potentiation. GRIPAP1 also serves as a signaling hub, participating in the activation of the JNK signaling cascade, and has further regulatory roles through posttranslational modifications. While not a classic drug target, its dysregulation is implicated in neurological disease processes and possibly oncogenic gene fusions in cancer[1][2][8][3].
