Group Y capsular polysaccharide of Neisseria meningitidis

Molecular classification
Polysaccharide antigen, Bacterial capsule component, Other (not a protein, enzyme, receptor or transporter; it is a complex carbohydrate)
01

Overview

The Group Y capsular polysaccharide of Neisseria meningitidis is a linear, high-molecular-weight carbohydrate polymer that composes the capsule surrounding Neisseria meningitidis bacteria of serogroup Y. It consists of alternating units of N-acetylneuraminic acid (sialic acid) linked to glucose via specific glycosidic bonds, forming a helical structure with distinct hydrophilic and hydrophobic surfaces. This capsule is the principal virulence determinant for the bacterium, allowing it to evade the host immune system, resist complement-mediated lysis, and cause invasive diseases such as meningitis and septicemia. The group Y polysaccharide is a key antigen in quadrivalent meningococcal conjugate vaccines (MenACWY). Detection of antibodies to this polysaccharide is used as a biomarker for vaccine-induced immunity.

Mechanism of action

Conjugate vaccines induce protective antibodies that recognize the group Y capsular polysaccharide, promoting bacterial clearance via opsonization and complement-mediated lysis

03

Biological functions

Immune evasion (prevents opsonization and lysis by host complement system)Major virulence factor (required for systemic invasive disease)Basis for vaccine antigenicity (elicits protective immune responses)
04

Disease associations

Infection (major determinant of Neisseria meningitidis pathogenicity, responsible for invasive meningococcal disease)
05

Safety considerations

Polysaccharide antigens alone can have limited immunogenicity in infants and young children, requiring conjugation to a carrier protein for effective vaccinationRare risk of immune escape through capsule switching or downregulationHypersensitivity to vaccine components (general vaccine concern)
06

Interacting drugs

Meningococcal conjugate vaccines containing group Y polysaccharide (e.g., MenACWY vaccines)
07

Biomarkers

Anti-group Y capsular polysaccharide IgG (serological marker for immunity and vaccine response)
