Growth arrest associated long non-coding RNA 1 (GASAL1) is an intergenic lncRNA located on chromosome 8q22.3, transcribed from the plus strand and comprised of four exons, with a mature length of 1,536 bases. GASAL1 expression is regulated by E2F transcription factors and acts as a negative regulator of cell cycle progression and cell proliferation, functioning as part of a feedback loop that restrains excessive E2F1 activity. Silencing GASAL1 enhances G1/S transition and increases cell proliferation, while overexpression inhibits these processes and sensitizes cells to apoptosis, particularly by modulating pRB phosphorylation and p21 expression. In vivo, loss of GASAL1 promotes tumor growth, and reduced levels are linked with poor prognosis in liver cancer. GASAL1 has been investigated for its roles in cancer development and as a potential biomarker for prognosis. No direct drug modulators are yet known, and the molecule is not a classical receptor or enzyme but belongs to the long non-coding RNA family.
