Growth differentiation factor 5 (GDF-5) is a secreted signaling protein belonging to the transforming growth factor-beta (TGF-β) superfamily, closely related to but distinct from the canonical bone morphogenetic proteins (BMPs)[1][3][5]. Synthesized as a precursor that is proteolytically cleaved, mature GDF-5 has a central role in skeletal and joint development, promoting the formation and repair of cartilage, bone, ligaments, and other musculoskeletal soft tissues[1][3][5]. It signals primarily through BMP type I (BMPR1b) and type II (BMPR2) receptors, activating downstream Smad proteins to control gene transcription[1]. In addition to its developmental and regenerative functions, GDF-5 also supports the survival of certain neurons, especially dopaminergic neurons, and recent research implicates it in energy homeostasis by promoting brown/beige adipocyte differentiation[2][4]. Mutations in the GDF5 gene cause a range of skeletal dysplasias and increase susceptibility to osteoarthritis and musculoskeletal degeneration, making it an active focus for tissue engineering and regenerative medicine[1][2][3][5]. Experimental therapies include gene delivery and recombinant protein administration, but GDF-5 is not yet an established drug target in routine clinical practice.
