Overview

This milieu includes growth factors such as Vascular Endothelial Growth Factor (VEGF) and Transforming Growth Factor-beta (TGF-β), which regulate cell proliferation and tissue repair, as well as cytokines like Tumor Necrosis Factor-alpha (TNF-α) and various interleukins that orchestrate immune and inflammatory responses (Nature Reviews Immunology, 2021). In pathological states like cancer or chronic inflammation, the composition of this milieu is often dysregulated, promoting disease progression, angiogenesis, and immune evasion (PubMed, 2022). While not a single therapeutic target, the individual components of the milieu are frequently targeted by monoclonal antibodies and small molecules to restore homeostatic signaling (StatPearls, 2023).
