Growth factor receptor-bound protein 10 (GRB10) is an adaptor protein crucial for intracellular signal transduction. It is defined by its interaction with receptor tyrosine kinases such as the insulin receptor and IGF1 receptor, where it generally acts to inhibit downstream signaling and cell proliferation. GRB10 binds targets via its SH2 and BPS domains and is involved in growth suppression, regulation of metabolic insulin action, energy metabolism, and neuronal development. Highly regulated by genetic imprinting, GRB10 has tissue- and isoform-specific roles. Disease associations include Silver-Russell syndrome, cancer, diabetes, and cardiovascular disease. Experimental evidence shows that disruption of GRB10 results in altered growth, metabolic disturbance, and behavioral changes in animal models[1][2][3][5][6].
