Overview

The growth hormone–releasing hormone receptor (GHRHR) is a G protein-coupled receptor (GPCR) that specifically binds growth hormone-releasing hormone (GHRH). Upon binding its ligand, GHRH, the receptor activates a stimulatory Gs protein, which in turn triggers the production of cyclic AMP (cAMP) via adenylate cyclase. Binding of GHRH to its receptor stimulates both synthesis and secretion of growth hormone (GH). Mutations in the GHRHR gene can cause isolated growth-hormone deficiency (IGHD), leading to short stature or Dwarfism of Sindh syndrome. Dysregulation or abnormal expression has been implicated in disorders involving GH excess or deficiency as well as some cancers due to aberrant signaling by certain splice variants.
