Overview

The Growth Hormone-Releasing Hormone Receptor (GHRHR) is a Class B G protein-coupled receptor primarily expressed in the anterior pituitary gland. It mediates the effects of growth hormone-releasing hormone (GHRH), stimulating the synthesis and release of growth hormone. GHRHR plays a critical role in growth, metabolism, and development. Mutations in GHRHR can lead to isolated growth-hormone deficiency (IGHD). Dysregulation or overexpression of GHRHR has been implicated in various cancers, making it a potential therapeutic target for antagonists.

Mechanism of action

Agonist binding activates Gs protein, leading to increased cAMP production and GH synthesis/release. Antagonists block GHRH binding, inhibiting downstream signaling.
