Overview

Growth hormone-releasing hormone receptor (GHRHR) mRNA is the transcript responsible for the synthesis of the GHRHR protein, a Class B G protein-coupled receptor primarily expressed on the surface of somatotroph cells in the anterior pituitary gland. The encoded receptor plays a critical role in the hypothalamic-pituitary-somatotropic axis by mediating the effects of hypothalamic GHRH, which triggers a signaling cascade via the Gs/adenylyl cyclase pathway to stimulate the synthesis and pulsatile secretion of growth hormone (GH) [UniProt: P35372]. Proper regulation of GHRHR mRNA expression and splicing is essential for normal somatotroph development and postnatal growth. Mutations in the GHRHR gene or defects in its mRNA processing are primary causes of Type IB isolated growth hormone deficiency (IGHD), a condition characterized by severe short stature and low but detectable GH levels [NCBI Gene: 2692]. While current clinical therapeutics like Tesamorelin and Sermorelin target the GHRHR protein to stimulate GH release, the mRNA transcript itself represents a potential target for emerging RNA-based therapies designed to modulate receptor density in growth-related disorders [PubMed: 10735251]. Monitoring serum IGF-1 and GH levels serves as the primary method for assessing the efficacy of interventions targeting this pathway.
