Overview

Growth hormone-releasing hormone receptor splice variant 1 (SV1) is a variant of the human growth hormone-releasing hormone receptor generated by alternative splicing. Unlike the canonical GHRH receptor, SV1 contains a unique N-terminal extracellular domain resulting from the replacement of the first three exons by a fragment from intron 3. SV1 is a functional G protein-coupled receptor, expressed in a spectrum of human cancers, where it mediates both ligand-dependent and ligand-independent cellular proliferation. SV1 displays biased intracellular signaling, preferentially recruiting β-arrestins rather than activating the classical G_s-cAMP pathway. This receptor is implicated in cancer progression, and antagonists targeting SV1 are under investigation for therapeutic interventions in multiple tumor types[1][2][5][4][7].
