Overview

GS1-594A7.3, also known as ACE2 divergent transcript (ACE2-DT), is a human-specific, independently transcribed long noncoding RNA (lncRNA) situated just upstream (within 722 base pairs) of the ACE2 gene on the X chromosome. The transcript is poorly conserved across vertebrate species and is primarily found in the nucleus of several human cell lines. Observations suggest that its expression frequently overlaps with ACE2 transcript levels in various tissues, and there is evidence for a shared or bifunctional promoter region. This close physical and regulatory relationship implies ACE2-DT may regulate ACE2 expression in *cis* (affecting neighboring ACE2 gene). While the detailed functions of ACE2-DT remain to be elucidated, it is hypothesized to modulate ACE2 expression levels, which could influence susceptibility to SARS-CoV-2 infection and COVID-19 pathology. There are currently no known drugs, clinical biomarkers, or safety concerns directly associated with ACE2-DT itself[2][5]. ACE2-DT is NOT a protein-coding target or a classical therapeutic target such as a receptor, enzyme, transporter, or ion channel. It is a long noncoding RNA whose interest lies in its possible regulatory effects on ACE2, a clinically relevant receptor for SARS-CoV-2. ACE2-DT is not currently the focus of direct drug targeting, and its functions are still under investigation, primarily using humanized mouse models[2][5].
