Overview

GTP-binding protein Rit2 is a neuron-specific small GTPase belonging to the Ras superfamily, principally regulating neuronal differentiation, survival, and signaling[1][2][5]. RIT2 is tightly expressed in various neuronal populations—including retinal, dopaminergic, and other neurons—in both the central and peripheral nervous system[2]. It interacts with the dopamine transporter and modulates its internalization in specialized membrane microdomains, impacting dopaminergic signaling, and is implicated in neurotrophin-mediated differentiation via activation of b-Raf and p38 MAP kinase signaling cascades[1][2]. RIT2 also modulates neuronal gene transcription through interactions with transcription factors such as Brn-3a/POU4F1[2][5]. Genetic studies link RIT2 to Parkinson’s disease risk and suggest involvement in schizophrenia and other neuropsychiatric disorders[2][3][5]. No approved drugs directly target RIT2; mechanisms for RIT2 modulation for therapeutic purposes remain an avenue for research.
