Overview

GTPase IMAP family member 1 (GIMAP1) is a small GTP-binding protein belonging to the immuno-associated nucleotide (IAN) subfamily and AIG1 domain-containing GTPases. It is preferentially expressed in lymphoid tissues, particularly in T and B lymphocytes throughout development, and localizes mainly to the endoplasmic reticulum and Golgi apparatus[1][2][4]. GIMAP1 is critical for the survival and maintenance of mature T and B cells; deficiency leads to peripheral lymphopenia and impaired immune function in animal models[2][4]. It is upregulated during Th1, but not Th2, cell differentiation, and has been associated with immune regulation and apoptosis, possibly functioning via pathways distinct from classical BCL-2 family proteins[1][2][4]. Aberrant expression of GIMAP1, including increased expression due to hypomethylation, is observed in certain lymphoid cancers, notably diffuse large B-cell lymphoma[2]. There are currently no known direct pharmacological modulators targeting GIMAP1, nor are drug mechanisms of action or safety concerns defined in the literature, but GIMAP1’s role in lymphocyte survival positions it as a potential therapeutic and biomarker candidate in immune and hematologic diseases[2][4].
