Overview

GTPase IMAP family member 8 (GIMAP8) is a member of the GIMAP family of small GTPase proteins, uniquely possessing three GTPase domains, which is exceptional among GIMAPs and small GTPases in general[2][4]. It is primarily expressed in hematopoietic and lymphoid tissues, including thymocytes and mature T cells[4]. GIMAP8 exerts anti-apoptotic effects within the immune system and is involved in the responses to infections, suggesting a key role in lymphocyte survival and homeostasis[3][1]. GIMAP8-deficient mice show normal T cell development but have reduced numbers of recirculating B cells in the bone marrow, implicating it in B cell maintenance[4]. The gene belongs to the AIG1 domain–containing protein family, which clusters on human chromosome 7q36.1[1][5]. Diseases associated with defects or alterations in GIMAP8 include hypoinsulinemic hypoglycemia with hemihypertrophy and hypogonadotropic hypogonadism with or without anosmia[1]. No specific drugs, mechanisms of drug action, or established biomarkers are currently linked to GIMAP8, and there are no documented safety concerns regarding therapeutic targeting of this protein[1][3][4].
