Overview

The O6 position of guanine refers to the oxygen atom attached to the sixth carbon of the guanine base in DNA. This site is a key target for alkylation and methylation damage, leading to mispairing with thymine during DNA replication and subsequent G:C → A:T transition mutations. It is primarily repaired by MGMT. Modification at this site is highly mutagenic and carcinogenic.
