The N7 position of guanine refers to the nitrogen atom at the seventh position on the guanine base, one of the four main nucleobases in DNA and RNA. This site is highly nucleophilic and is a primary target for alkylation and adduct formation by various chemicals, including carcinogens and certain chemotherapeutic agents. N7-guanine adducts are frequently used as biomarkers to assess exposure to genotoxic agents. Methylation at this site also occurs naturally during mRNA capping (m^7G cap), which is essential for mRNA stability and translation initiation in eukaryotes.
