Overview

The N7 position of the guanine base within nuclear double-stranded DNA is a critical nucleophilic site and a primary target for various chemotherapeutic agents. Due to its high electron density and accessibility within the major groove of the DNA helix, it is the most reactive position for electrophilic attack by alkylating agents and platinum-based coordination complexes (StatPearls, 2023). When drugs like cisplatin or cyclophosphamide bind to the N7 atom, they form stable covalent adducts that can further evolve into interstrand or intrastrand cross-links (PubMed, PMID: 11483377). These cross-links physically distort the DNA structure, effectively blocking the progression of DNA polymerase and RNA polymerase, thereby inhibiting DNA replication and transcription (NIH, PubChem). The resulting persistent DNA damage activates cellular stress responses and DNA damage checkpoints, ultimately triggering programmed cell death (apoptosis) in rapidly dividing cancer cells. While highly effective in oncology, the non-specific nature of these interactions leads to damage in healthy tissues, resulting in significant side effects such as bone marrow suppression and the potential for long-term mutagenic effects (Wikipedia, Alkylating antineoplastic agent).
