Overview

Guanine nucleotide-binding protein-like 1 (GNL1) is an evolutionarily conserved putative nucleolar GTPase of the HSR1_MMR1 GTPase subfamily, predominantly localized in the nucleolus with nuclear-cytoplasmic shuttling that's dependent on the cell cycle. GNL1 is upregulated in several human cancers and promotes cell proliferation and survival, at least in part, by activating the AKT/p21 signaling pathway, stabilizing cytoplasmic p21, inhibiting apoptosis via modulation of Bcl2 family proteins and caspases, and enhancing cell cycle progression through effects on pRb phosphorylation and G1/S and G2/M phase transitions. Increased GNL1 correlates with poor clinical outcome in some cancer types and suggests involvement in tumorigenesis. There are currently no reported drugs or inhibitors specifically targeting GNL1 in clinical use or development.
