Overview

Guanine nucleotide-binding protein subunit alpha-14 is a member of the G protein alpha-q family (GNA14), functioning as part of the heterotrimeric G protein signaling complex. Upon activation of cell surface GPCRs, GNA14 facilitates GDP-GTP exchange and activates phospholipase C-beta, triggering hydrolysis of membrane phospholipids and release of intracellular messengers such as diacylglycerol (DAG) and inositol trisphosphate (IP3), resulting in activation of protein kinase C and calcium signaling. GNA14 is implicated in diverse physiological processes including platelet aggregation, cardiovascular regulation, glucose metabolism, and cell proliferation. It may play a role in disease states such as vascular tumors and cardiomyopathies, and pharmacological inhibition has demonstrated therapeutic potential in preclinical models.
