Overview

Guanine nucleotide-binding protein subunit beta-4 (GNB4) is a protein coding gene that encodes the beta subunit of heterotrimeric G proteins. These proteins are membrane-bound GTPases that integrate signals between cell surface receptors (especially G protein-coupled receptors) and effector proteins, playing essential roles in transmembrane signaling. The beta subunit regulates the activity of alpha subunits and modulates signal transduction through interaction with receptors and effectors. Mutations in GNB4 have been linked to inherited neurological disorders such as Charcot-Marie-Tooth disease and have also been implicated in cancer progression. The beta subunit acts in concert with its gamma partner to facilitate receptor-triggered GDP–GTP exchange and downstream signaling events.
