Overview

KRAS is a small GTPase that acts as a molecular switch in cell signaling pathways, particularly the RAS/MAPK pathway. It regulates cell proliferation, differentiation, and survival by cycling between an inactive GDP-bound state and an active GTP-bound state. Mutations in KRAS, especially at codons 12, 13, and 61, are common in various cancers and often lead to constitutive activation of the protein, driving uncontrolled cell growth. Direct targeting of KRAS has been historically challenging, but covalent inhibitors for specific mutants like KRAS(G12C) have been developed. Newer approaches include allosteric inhibitors and PROTACs.
