Guanylate-binding protein 3 (GBP3) is an interferon-inducible large GTPase belonging to the guanylate-binding protein family. It binds and hydrolyzes guanine nucleotides (GTP, GDP) and plays a crucial role in innate immunity against viruses, bacteria, and protozoa. GBP3 is part of host defense mechanisms, promoting the lysis of pathogen-containing vacuoles, facilitating the release of pathogenic ligands into the cytosol, and positively regulating inflammasome assembly. Its expression and mutational profiles are associated with the prognosis and immune landscape in multiple human cancers, where it may influence immune cell infiltration and cancer progression. The GBP3 protein shares homology with other GBPs, features conserved GTPase motifs, and is regulated by interferons as part of the antiviral response.
