Guanylate-binding protein 4 (GBP4) is a member of the guanylate-binding proteins, a subset of the dynamin superfamily of large GTPases that are induced by interferons and play critical roles in innate immune defense, particularly via cell-autonomous immunity against a range of intracellular pathogens including bacteria, viruses, and protozoa. Like other GBPs, GBP4 binds and hydrolyzes GTP, and is involved in processes such as autophagy, inflammasome activation, and potentially membrane remodeling and restriction of pathogen replication. While much research has focused on other family members (like GBP1, GBP2, and GBP5), GBP4 shares significant sequence and structural similarity, and is presumed to act by analogous mechanisms. As of now, GBP4 is not directly targeted by approved drugs but its expression is a marker of interferon pathway activation, and modulation of its activity could impact immune responses to infection and inflammation.
