Overview

Guanylate cyclase 1 soluble subunit alpha-1 (GUCY1A1) is the alpha subunit of the soluble (cytosolic) guanylate cyclase enzyme complex. This enzyme is a heterodimer composed of alpha and beta subunits, acting as the primary receptor for nitric oxide (NO) in human cells. Upon binding NO, the active enzyme catalyzes the conversion of GTP to cyclic GMP (cGMP), a crucial signaling molecule involved in regulating vascular tone, platelet aggregation, and cellular proliferation. Mutations in GUCY1A1 have been implicated in Moyamoya disease and other vasculopathies. Approved drugs such as riociguat and vericiguat target this enzyme to promote vasodilation in pulmonary hypertension and heart failure.
