Guided entry of tail-anchored proteins factor 1 (GET1) is a membrane protein of the endoplasmic reticulum (ER) that is essential for the post-translational delivery and insertion of tail-anchored (TA) proteins into the ER membrane. It acts as a membrane receptor, together with CAMLG/GET2, for the cytosolic factor GET3/TRC40, which recognizes and binds the transmembrane domains of TA proteins. GET1 ensures proper topology and efficient ER insertion of its cargo and is part of a conserved system required for targeting proteins critical in vesicle trafficking, apoptosis, and organelle biogenesis. Diseases associated with mutations or dysregulation of GET1 include congenital heart disease, and its genomic locus has relevance in Down syndrome pathology[2][3][6][7].
