Overview

Guided entry of tail-anchored proteins factor 4 (GET4) is a highly conserved cytoplasmic protein that is part of the GET (guided entry of tail-anchored proteins) pathway, which is responsible for the post-translational targeting of tail-anchored (TA) membrane proteins to the endoplasmic reticulum (ER)[1][2][3]. GET4 forms a stable complex with GET5 (Get4/5 complex), acting as an intermediary between the ribosome and the targeting factor GET3. GET4 binds to GET3 in a nucleotide-dependent manner and, together with GET5, facilitates the loading of TA substrates onto GET3, priming it for membrane targeting. Structurally, GET4 is composed of α-helical domains with regions resembling TPR-like motifs, although it lacks classical sequence motifs[1][3][4]. While essential for correct cellular protein trafficking, GET4 is not a canonical therapeutic target (such as a receptor, enzyme, or transporter), nor are there any known drugs that modulate its function in clinical or experimental settings[2][3][4]. Its dysfunction can cause defects in the delivery of tail-anchored proteins, which may disrupt membrane protein homeostasis in eukaryotic cells[2].
