Target intelligence / Profile preview
The gut microbiota and gastrointestinal mucosal barrier constitute a dynamic interface critical for maintaining systemic homeostasis and immune tolerance (Frontiers in Immunology, 2021). The mucosal barrier serves as a multi-layered defense system, comprising a biochemical mucus layer, a physical epithelial cell layer joined by tight junctions, and an underlying mucosal immune system (Nature Reviews Gastroenterology & Hepatology, 2023). The gut microbiota interacts with this barrier by producing metabolites like short-chain fatty acids (SCFAs) that nourish colonocytes and strengthen junctional complexes (Cell Host & Microbe, 2020). Disruptions in this relationship, characterized by microbial dysbiosis and increased intestinal permeability, are central to the pathogenesis of inflammatory bowel disease (IBD), metabolic disorders, and certain systemic autoimmune conditions (Science, 2019). Pharmacological strategies targeting this axis include the use of GLP-2 analogs like Teduglutide to promote epithelial growth, zonulin inhibitors like Larazotide to stabilize tight junctions, and various biotherapeutics designed to restore microbial diversity (Journal of Clinical Investigation, 2022).
Modulation of microbial diversity, enhancement of epithelial tight junction integrity, and regulation of mucosal immune signaling.
