H2A clustered histone 4 (H2AC4) is a core histone protein in eukaryotes responsible for forming the nucleosome structure of chromatin by helping package DNA. It is one member of a large family of H2A histone variants that play a central structural role in chromatin by forming an octameric nucleosome core with other histones (H2B, H3, H4)[1][4]. H2A clustered histone 4 is encoded by replication-dependent histone genes clustered in the genome and possesses subtle sequence variations, especially in the C-terminus, that influence how DNA is packed and accessed. While not a typical drug target or receptor, alterations in expression or modification of H2A histones can be linked to changes in chromatin architecture and gene regulation, and somatic mutations or aberrant post-translational modifications may contribute to cancer epigenetics and other chromatin-related disorders[1][3][4].
