Haematopoietic cell kinase (HCK) is a non-receptor tyrosine kinase belonging to the Src family of kinases. It is primarily expressed in cells of the myeloid lineage (such as monocytes, macrophages, and neutrophils) and B-lymphocytes. HCK plays a central role in regulating immune cell signaling, proliferation, migration, adhesion, and inflammatory responses. Excessive activation or aberrant expression of HCK is linked to several leukemias as well as solid tumors such as breast and colon cancer. Selective inhibition of HCK is under investigation for cancer therapy due to its dual role: directly suppressing malignant cell growth/proliferation; indirectly reducing pro-tumorigenic changes within the microenvironment via effects on immune cells/macrophages.
