Overview

Haeme-mediated free radical generation is a critical biochemical process in Plasmodium species. It involves the release of free heme during hemoglobin digestion, leading to ROS production via Fenton chemistry. This process influences pathogenesis and serves as a target for antimalarial therapies. Plasmodium converts toxic free heme into hemozoin to mitigate oxidative stress. Many antimalarial drugs, such as artemisinin and chloroquine, exploit this pathway to induce parasite death. Excessive free heme released during hemolysis can also trigger inflammation and thrombosis in the host.

Mechanism of action

Artemisinin derivatives interact with ferrous iron in heme to generate cytotoxic free radicals. Chloroquine inhibits the biocrystallization of hemozoin from free heme.
