The Haemophilus influenzae type b (Hib) capsular polysaccharide-protein conjugate is the active component in Hib conjugate vaccines, designed to prevent invasive diseases caused by H. influenzae type b bacteria. The vaccine consists of the polyribosylribitol phosphate (PRP) capsule polysaccharide conjugated to a carrier protein to enhance immunogenicity, particularly in infants. Common carrier proteins include tetanus toxoid, diphtheria toxoid, Neisseria meningitidis outer membrane protein, and diphtheria mutant carrier protein CRM197. The conjugation process enables the immune system to recognize the PRP antigen as a T-cell dependent antigen, leading to robust antibody production and immunological memory against Hib bacteria. It is administered intramuscularly as part of routine childhood immunization schedules.
