Polyribosylribitol phosphate (PRP) is the capsular polysaccharide antigen specific to Haemophilus influenzae type b (Hib). It is a high-molecular-weight polymer that forms the outer capsule of Hib bacteria, serving as a major virulence factor and the primary target for protective immune responses and vaccine development. PRP consists of repeating units composed of D-ribose, ribitol, and phosphate. It protects Hib from host immune defenses by inhibiting complement-mediated phagocytosis and shielding bacterial surface antigens. Antibody responses against PRP are critical for clearance of Hib bacteria. PRP is used as a conjugate with carrier proteins in vaccines to elicit robust T-cell-dependent immunity.
