The Haemophilus influenzae type b (Hib) polysaccharide capsule is a critical virulence factor that enables the bacterium to evade host immune responses and cause invasive diseases. It is composed of polyribosyl-ribitol-phosphate (PRP). The PRP component forms the basis for current conjugate vaccines against Hib, which induce strong T-cell dependent immune responses. The capsule protects Hib from phagocytosis and complement-mediated killing, making it the main determinant for virulence.
Vaccines induce antibody production against PRP, leading to opsonization and complement-mediated killing of Hib.
