Overview

The hair follicle dermal papilla (DP) is a compact cluster of specialized mesenchymal fibroblasts forming the core mesenchymal niche at the base of the hair follicle, in direct contact with epithelial progenitors. It orchestrates cyclic hair regeneration by secreting signaling molecules like Wnt activators, BMP inhibitors, and FGF ligands to control stem cell quiescence in telogen, activation in hair germ, and proliferation/differentiation in the matrix during anagen.[1][5] DP size and signaling strength determine hair follicle size, type, and cycling frequency, with ablation studies confirming its necessity for de novo hair growth but not epithelial maintenance.[1] In disease, DP dysfunction contributes to androgenetic alopecia through altered testosterone processing and signaling imbalances.[3] Emerging therapies explore transplanting exogenous DP cells (DPCs) or dermal sheath cells, which home to niches via CXCL13/CXCR5 to promote thicker hair shafts and prolonged anagen, though challenged by immune issues and homing efficiency.[2] The DP interacts dynamically with surrounding dermal sheath and adipocytes, highlighting its role as an instructive niche for epithelial-mesenchymal crosstalk essential for lifelong hair production.[1][5]
