Overview

Hair follicle stem cell (HFSC) activation refers to the process by which normally quiescent stem cells residing in the hair follicle bulge become activated to proliferate and initiate a new cycle of hair growth. This activation is a tightly regulated event, integrating both intrinsic cellular mechanisms and extrinsic signals from the surrounding microenvironment. HFSCs are essential for cyclical regeneration of hair follicles, fueling new hair growth during the anagen (growth) phase. They also contribute to skin wound healing. Key pathways involved include Wnt/β-catenin (activation), BMP (quiescence), FGF7 (activation), and JAK/STAT. Metabolic regulation, particularly glycolytic metabolism and lactate production, is critical for HFSC activation. The niche and microenvironmental influence, including macrophages, also play significant roles. Intrinsic regulatory mechanisms involve transcription factors and epigenetic regulation.
