Hairy and enhancer of split 6 (HES6)

Hairy and enhancer of split 6 (HES6) is a basic helix-loop-helix transcription cofactor that regulates cell differentiation in multiple cell types by antagonizing transcriptional repressors like HES1. It does not bind DNA directly, but modulates gene expression by suppressing HES1-mediated transcriptional repression and promoting the differentiation of neurons. HES6 interacts with other transcription factors through a specific tetrapeptide domain at its C-terminus and integrates signals from key developmental pathways, such as Notch signaling. Diseases associated with HES6 include brain cancer and Alagille syndrome. HES6 is not an established therapeutic target, and no drugs directly interact with it; its role is primarily in the regulation of transcription, cell fate, and differentiation during development and carcinogenesis.
