Overview

HAND2 antisense RNA 1 (HAND2-AS1) is a human long non-coding RNA (lncRNA) gene located on chromosome 4 (sometimes stated as chromosome 2, but most current databases place it on chromosome 4)[1][7][8]. HAND2-AS1 is transcribed from the antisense strand relative to the HAND2 gene and is highly expressed in the heart, as well as in select other tissues—notably neuroblastoma cells, where its expression is often dysregulated[1]. The primary function of HAND2-AS1 is believed to be the regulation of gene expression, partly through acting as a molecular "sponge" for microRNAs, thereby modulating the availability of miRNAs to repress their mRNA targets[1][8]. This regulatory role impacts cellular proliferation, differentiation, apoptosis, and organ development—particularly in the heart and nervous system[1][2][8]. HAND2-AS1 has been implicated in the development and progression of various cancers (including neuroblastoma) and in cardiovascular pathology, where it may influence cardiac right ventricle formation[1][8]. It is not a conventional therapeutic target such as a receptor, enzyme, transporter, or ion channel, and no drugs are documented to directly interact with or modulate HAND2-AS1 as of now. However, HAND2-AS1’s dysregulation in disease and its ability to influence cell fate make it a subject of active research in both basic biology and potential future medical applications[1].
