Hantaan virus envelope glycoprotein Gn is a critical structural component of the Hantaan virus, which is the primary cause of Hemorrhagic Fever with Renal Syndrome (HFRS) in Eurasia [3, 11]. It is encoded by the M segment of the viral genome and undergoes post-translational cleavage to form a mature protein that assembles into heterotetrameric spikes with the Gc glycoprotein on the viral envelope [2, 8]. These spikes are responsible for the initial attachment of the virus to host cell receptors, including β3 integrins and C1QBP, thereby facilitating viral entry via clathrin-mediated endocytosis [2, 15]. Gn also plays a significant role in the assembly and budding of new virions by interacting with the viral nucleocapsid protein [2, 3]. As the primary target for the host's neutralizing antibody response, Gn is a major focus for the development of vaccines and therapeutic monoclonal antibodies [11, 14]. Experimental treatments, such as the monoclonal antibodies HTN-Gn1 and MIB22, aim to neutralize the virus by blocking its ability to infect host cells [14, 15].
