Overview

The Hantavirus nucleocapsid protein (N protein) is the primary structural component of the hantavirus virion and plays a central role in the viral life cycle [UniProt, 2024]. It is responsible for encapsidating the three segments of the negative-sense RNA genome, forming the ribonucleoprotein (RNP) complex required for replication and transcription [PubMed, 2019]. Beyond its structural role, the N protein interacts with host cell factors to modulate the immune response and facilitate viral assembly [NCBI, 2023]. Because it is the most abundant protein produced during infection and is highly conserved across different hantavirus species, it is the dominant antigen for B-cell and T-cell recognition [Lindgren et al., 2021]. This makes the N protein's conformational and linear epitopes essential targets for diagnostic assays and potential vaccine candidates [Frontiers in Microbiology, 2019]. While current antiviral treatments like Ribavirin primarily target the viral polymerase, research into monoclonal antibodies and small molecules that specifically disrupt N protein function or its interaction with RNA is ongoing [Journal of Virology, 2020].
