Overview

Harbinger transposase-derived nuclease (HARBI1) is a protein encoded by the HARBI1 gene in humans, functionally classified as a domesticated transposase-derived nuclease[2][7]. HARBI1 is thought to have nuclease activity and metal ion binding ability based on sequence homology and in vitro assays, but it does not have transposase activity[2]. It localizes to centriolar satellites, cytosol, and the plasma membrane[2][7]. HARBI1 is most closely related to the PIF/Harbinger families of DNA transposons. In mammals and vertebrates, the gene is believed to have evolved (domesticated) from an ancient animal transposase, with no current transpositional activity[1][5]. In cell studies, HARBI1 can physically interact with NAIF1, another domesticated transposon-derived protein. This interaction is thought to have a role in nuclear import and a hypothetical DNA recombination process, but the precise biological function in humans remains unclear[1]. While associated via genetic studies with rare disease phenotypes, there is no established evidence for HARBI1 as a pharmacological or clinical target, nor is it known to be modulated by drugs or used as a biomarker[2][7].
