Overview

HAUS augmin-like complex subunit 6 (HAUS6) is a core component of the hetero-octameric augmin complex, which is essential for branching microtubule nucleation during cell division[1]. The augmin complex acts as a critical recruitment factor, localizing the γ-tubulin ring complex (γ-TuRC)—the major microtubule nucleator—to pre-existing microtubules. This process is fundamental for spindle assembly during mitosis and meiosis. HAUS6 contains a highly conserved calponin homology (CH) domain that binds directly to the inter-protofilament groove of microtubules via a basic surface, anchoring the augmin complex and determining the orientation and angle of microtubule branching[1]. The interaction is essential for the correct assembly and length of the mitotic spindle; loss or mutation of HAUS6 results in defective spindle formation and impaired cell division[1]. HAUS6, together with related subunits (HAUS2, HAUS7, HAUS8), forms the "TII subcomplex" of augmin, responsible mainly for microtubule binding, while the "TIII subcomplex" interfaces with γ-TuRC[1][2][3]. There are no known drugs that selectively target HAUS6, nor is it currently considered a direct therapeutic target, but defects in augmin function have been implicated in genomic instability associated with cancer[1].
