Overview

HAUS augmin-like complex subunit 8 (HAUS8) is a microtubule-associated protein and an essential component of the eight-subunit augmin (HAUS) complex, which is crucial for mitotic spindle assembly, the maintenance of centrosome integrity, and the organization of microtubules during mitosis and cytokinesis. HAUS8 interacts directly with spindle microtubules, helps recruit the γ-tubulin ring complex, and enables microtubule branching from pre-existing spindle fibers. It contains a monopartite nuclear localization signal (NLS) motif that mediates competitive interactions with both microtubules and importins, regulated by the Ran-GTP pathway and by phosphorylation. Genetic or functional deficiency of HAUS8 disrupts mitotic spindle formation, promotes chromosome instability, and impairs cell division. Beyond its cytoskeletal roles, HAUS8 can also modulate innate immune signaling by interacting with the VISA complex to promote RLR-VISA-dependent antiviral signaling, suggesting possible links between cytoskeletal integrity and immune response. While it has been identified as a cancer-associated antigen (NY-SAR-48), HAUS8 is not currently targeted by known drugs, nor is it classified as a classical therapeutic target such as a receptor, enzyme, or transporter.
