Overview

HCLS1-binding protein 3 (HS1BP3) is a human protein encoded by the HS1BP3 gene on chromosome 2p24.1. It is an adaptor/scaffold protein that interacts with the hematopoietic lineage cell-specific protein HS1 via SH3 domains, suggesting a role in modulating lymphocyte activation and intracellular signaling pathways such as IL-2 signaling. HS1BP3 also binds members of the 14-3-3 protein family—prominent in neuronal cells—and may regulate apoptosis and stimulate AP-1-mediated transcription. While mutations in HS1BP3 have been connected to hereditary essential tremor, direct involvement in other diseases is not well-established, and no drugs currently target HS1BP3 for therapeutic intervention.
