Overview

Heart- and neural crest derivatives-expressed protein 2 (HAND2) is a basic helix-loop-helix transcription factor that orchestrates critical developmental pathways, particularly in the embryonic heart, limb buds, craniofacial structures, and neural crest derivatives. It promotes right ventricular morphogenesis, vascularization, and cardiac neural crest development, and is crucial for proper limb and craniofacial patterning. HAND2 functions both by direct DNA binding at consensus E-box sequences and through DNA-binding-independent mechanisms, forming multiprotein transcriptional complexes. Its tightly regulated expression involves chromatin modification and microRNA control; disruption of HAND2 leads to congenital heart defects, craniofacial malformation, impaired limb and neural differentiation, and may influence cancer cell plasticity via mesenchymal–epithelial transitions. HAND2 is emerging as both a research and therapeutic target, particularly for cardiac regeneration and developmental syndromes.
