Heart spirit, translated from the Traditional Chinese Medicine (TCM) term 'Xin Shen,' is a conceptual therapeutic target representing the integration of cardiovascular health and neuropsychiatric stability. In TCM theory, the Heart is the primary organ that houses the 'Spirit,' which is responsible for governing consciousness, emotions, and the sleep-wake cycle; disturbances in this state, often called 'Heart spirit disquiet,' manifest as clinical symptoms such as anxiety, insomnia, and palpitations [1, 2]. While Heart spirit is not a singular molecular entity like a specific receptor or enzyme, contemporary network pharmacology research identifies it as a multi-target locus for traditional medicines [7, 11]. Research indicates that 'soothing' or 'stabilizing' the Heart spirit involves modulating various molecular pathways, including the serotonergic system (specifically HTR2A), SIRT1-mediated mitochondrial biogenesis, and cardiac voltage-gated ion channels [14, 20]. Consequently, it serves as a functional target for addressing disorders of the heart-brain axis through multi-component pharmacological interventions [1, 13].
Modulation of Heart spirit involves multi-target pharmacology, including the stabilization of voltage-gated sodium and potassium channels to prevent arrhythmia, regulation of the 5-HT and GABAergic neurotransmitter systems to alleviate anxiety and insomnia, activation of SIRT1-mediated mitochondrial biogenesis, and inhibition of neuroinflammatory cytokines such as TNF-α and IL-6 [1, 7, 14, 20].
