Overview

HEAT repeat-containing protein 5A (HEATR5A) is a large, highly conserved protein containing HEAT repeat structural motifs, which are composed of tandem pairs of alpha helices linked by short loops[1][3]. HEATR5A is part of the alpha solenoid protein family, whose characteristic architecture facilitates multiple protein–protein and protein–nucleic acid interactions[1]. In mammals, HEATR5A localizes to the trans-Golgi network and endosomes and is thought to play a major role in membrane trafficking, including intra-Golgi recycling and the recruitment and localization of the adaptor protein complex-1 (AP1)[2]. HEATR5A forms at least two biochemically distinct complexes with mutually exclusive binding co-factors, reflecting potential multifunctionality[2]. To date, HEATR5A is not established as a therapeutic target, and there are no approved drugs or known ligands associated with it. Its dysfunction in related family members can result in severe cellular or developmental phenotypes, but no direct links to specific human diseases or clinical biomarker roles have been established for HEATR5A itself[2].
