Overview

Heat shock factor 1 (HSF1) is a highly conserved transcription factor that serves as the primary regulator of the heat shock response (HSR), orchestrating cellular defense mechanisms against proteotoxic stress. Upon activation by stress, HSF1 trimerizes, translocates to the nucleus, and binds to heat shock elements (HSEs) to upregulate the expression of heat shock proteins (HSPs) and other cytoprotective genes involved in protein homeostasis and cell survival. HSF1 is implicated in various diseases including neurodegenerative disorders, cancer, and aging.

Safety considerations

Potential for off-target effects due to broad influence on gene expression.Context-dependent regulation; activation may have variable effects depending on stress type and duration.Overactivation may contribute to cancer progression and drug resistance.
