Heat shock protein–peptide complexes are molecular assemblies where heat shock proteins (such as HSP70 and HSP90) bind to peptide fragments derived from self, stress-induced, or tumor proteins[4][5]. These complexes play crucial roles in preventing protein misfolding and aggregation by acting as chaperones—folding or refolding misfolded proteins under cellular stress conditions[1][2][3]. In addition to their intracellular roles, these complexes can be found extracellularly (especially in cancer and infection), where they serve as antigen carriers, facilitating the uptake, processing, and cross-presentation of antigenic peptides to the immune system, and triggering adaptive immune responses[4][5][6]. Because of these properties, HSP–peptide complexes are employed experimentally in cancer immunotherapies and vaccine development[4][5]. They are considered promising therapeutic targets both for their protein quality control functions and immune stimulatory capacity, but current clinical approaches face safety and standardization challenges[4][5].
