The Heat shock protein 110–glycoprotein 100 (Hsp110–gp100) complex is a therapeutic chaperone-antigen construct primarily utilized in the development of cancer immunotherapies for melanoma. In this complex, Hsp110 (HSPH1) serves as a powerful molecular chaperone and adjuvant that stabilizes the melanoma-associated antigen gp100 (PMEL), preventing its aggregation and enhancing its delivery to antigen-presenting cells (APCs) such as dendritic cells. Upon uptake, Hsp110 promotes the cross-presentation of gp100-derived peptides on MHC class I molecules, which is critical for the activation of cytotoxic T lymphocytes (CTLs) capable of recognizing and destroying melanoma cells. Research, particularly from institutions like Roswell Park Comprehensive Cancer Center, has demonstrated that this complex is significantly more effective at eliciting anti-tumor immunity than the antigen alone. Clinical and preclinical studies focus on its ability to break immune tolerance to self-antigens in the context of advanced malignancy.
The complex acts as a chaperone-based vaccine where Hsp110 facilitates the uptake, processing, and cross-presentation of the gp100 antigen by dendritic cells to CD8+ T cells, thereby inducing a potent anti-tumor immune response.
